The relation to 'act' is real, but the common meaning 'respond' is not fully predictable for learners from the usual elementary sense of 're-' as 'again'.

Etymology

The word react comes from the parts re- meaning 'again' and act meaning 'do', the same root you see in action. So to react is to 'do' something in answer to what happens, in other words to respond.
