The cinema sense is conventional and partly guessable from mega-, but plex is not a transparent standalone English element for most learners.
Etymology
The word megaplex is like multiplex but bigger: mega- means 'large' and plex (as in multiplex) means 'many screens', so a megaplex is a very large movie building with many screens showing different films at the same time.
