The meaning 'a holiday honoring mothers' is directly and transparently derived from combining 'mother(s)' (a mother) with 'day' (a day set aside), i.e. a day for mothers. This compositional pattern is cross-linguistically common (e.g. Spanish Día de la Madre, French Fête des mères), so a B1 learner who knows the constituents will readily infer the meaning.
Etymology
Mother's Day may come from one woman's idea in the early 1900s, when she wanted one special day to thank her mother and asked people to give flowers and letters. That's why Mother's Day today is a holiday 'to honor mothers'.
